IRS990/Desc0Research Communities and Challenges: Sage Bionetworks conducts research independently as well as with collaborators worldwide across a variety of disease areas. We work to redefine how complex biological data is gathered, shared and used through open systems, incentives, and norms. Our work includes the building of platforms and services and undertaking research developing predictors relating to health. In 2015, Sage was engaged in over 30 active federal research projects, and nearly 50 projects overall. Research Communities active in 2015 included the following: The AMP AD Knowledge Portal is a premier repository and workspace for multi-omic data generated through the AMP-AD consortia - a public private partnership, with the goal of accelerating the development of new drugs for the treatment and prevention of Alzheimer's disease. Data was first publically released starting in March 2015. The Colorectal Cancer Consortium is a collection of colorectal cancer researchers and data scientists from 15 academic, pharmaceutical, and non-profit partners, which represents the first effort to generate consensus cancer, subtypes, taking a major step toward precision medicine in CRC. The CommonMind Consortium is a Public-Private Pre-Competitive partnership with the goal to generate and analyze large-scale genomic data from human subjects with neuropsychiatric disease and to make this data and the analytical results broadly available. March 2015 marked the first data release. The Infectious Disease Resilience project is designed to identify common characteristics of individuals that are resilient to infectious disease as a mechanism to understand the underlying biology. The mPower Public Researcher Portal is the data access point for the mobile app-based study piloting new approaches to monitoring key indicators of Parkinson Disease progression and diagnosis. The Progenitor Cell Biology Consortium (PCBC) is an NIH sponsored effort to identify and characterize progenitor cell lineages and to develop new strategies related to the transplantation of these cells. The goal of the PsychENCODE Consortium is a public, comprehensive, and integrated resource of human brain data with a focus on autism, mood disorders, and psychoses. Data generated by the Consortium will be disseminated to the public starting in 2016. The Synodos Consortium is a first-of-its-kind Nuerofibromatosis research collaboration dedicated to defeating the rare genetic disorder neurofibromatosis type 2. With data from 12 cancers The Cancer Genome Atlas (TCGA) is an effort to simultaneously analyze molecular platforms for patterns across cancer types. Working in Synapse this group created a common data resource and made results available through 18 published papers. In 2013, Sage partnered with DREAM (http://www.the-dream-project.org/), to host open computational challenges designed and run by a community of researchers from a variety of organizations, and with a solver community of more than 15,000 individuals. DREAM Challenges invite participants to propose solutions to fundamental questions about systems biology and translational medicine - fostering collaboration and building communities in the process. Sage's Challenges leverage the "wisdom of the crowd" to find new and better computational models and make these methods available to all. Active Challenges in 2015 included ALS Stratification Prize4Life DREAM Challenge - focuses on predicting the progression and survival of ALS patients. This effort will also work to significantly reduce the costs of future ALS clinical trials, aid clinical care and identify new disease predictors that can lead to novel biomarker development. AstraZeneca Drug Combination Prediction DREAM Challenge - designed to explore fundamental traits that underlie effective combination treatments and synergistic drug behavior using baseline genomic data. IRS990/MissionDesc0Sage Bionetworks is a strategic research organization with a mission to coordinate and link academic and commercial biomedical researchers, patients and citizens through a Commons that represents a new paradigm for genomics intellectual property, researcher cooperation and contributor evolved resources. This mission has several interdependent themes, including: conducting research on network models of disease; building the computational platform for the Commons; and activating public engagement. Sage Bionetworks is a non-profit research organization that seeks to develop predictors of disease and accelerate health research through the creation of open systems, incentives, and standards. We create strategies and platforms that empower researchers to share and interpret data on a colossal scale, crowdsource tests for new hypotheses, and contribute to knowledge through community challenges.

IRS990/ProgSrvcAccomActy2Grp/Desc0Sage Bionetworks Mobile Health (mHealth): In 2013, Sage expanded efforts in citizen engagement with the development of Bridge, a cloud-based collaborative science platform designed to incorporate open data, patient wisdom and public involvement into biomedical research. Bridge is a key piece of IT infrastructure essential for empowering citizens to share and track their data, to partner on research and to fund the projects that matter most to them. On Bridge, citizens, patients and researchers are able to use online tools that connect people, their data and their stories to build online communities focused on defining the disease or health research that matters most to patients and their families. With the 2015 launch of Apple's open source ResearchKit platform, Sage Bionetworks has been an integral partner in the design, development and launch of smartphone-enabled clinical studies across multiple disease areas. mHealth services include: software engineering for app design and development, data governance for e-consent and IRB submission and IRB-approved cloud-based infrastructure (Bridge Server) for study data collection, de-identification and hosting. In 2015, Sage launched the first two mobile app-based research studies on Bridge as part of Apple's ResearchKit. Enrollment in the Parkinson's mPower Study and the Share the Journey Breast Cancer Survivor Study opened in March 2015. The Parkinson's mPower Study uses microphone, accelerometer, and touch-screen sensors in a mobile app to measure fluctuations in gait, posture, voice, and manual dexterity, and explores correlations with environmental factors such as sleep and exercise. The Share the Journey Breast Cancer Survivor Study is designed to look at the long-term side effects of chemotherapy on mood, cognitive function, and energy level, exploring the intersection of these experiences with environmental factors to learn how symptoms can be more effectively managed. Active Challenges in 2015 included:ALS Stratification Prize4Life DREAM Challenge - focuses on predicting the progression and survival of ALS patients. ICGC-TCGA Somatic Mutation Calling -Tumor Heterogeneity DREAM Challenge - an international effort to improve standard methods for subclonal reconstruction to quantify and genotype each individual cell population present within a tumor.
