Baltimore Under Ground Science Space, Inc.'s (BUGSS) mission is to foster a community of citizen scientists, who can explore biotechnology in a safe, constructive and socially-responsible manner. BUGSS is a community laboratory that promotes education and understanding of biotechnology, doing so by offering courses, informational talks, community projects, workshops, student science-fair assistance and an open laboratory, where individuals can conduct biotechnology experiments. At BUGSS, we believe that biotechnology and synthetic biology are the fabrication technologies of the future, and a key component of a sustainable economy in the mid-Atlantic region. Governance Explanations

Form 990, Part VI, Section B, Line 11B

A copy of the draft 2020 Form 990 and supporting schedules was reviewed by officers and the Finance Committee in the second quarter of 2021, followed by a review and sign-off by the remaining members of the Board of Directors in the same quarter. All parties reviewed the data and had an opportunity to offer refinements to narrative language, as well as the overall presentation. The independent accountant also reviewed these forms. The final 2020 Form 990 and supporting schedules were electronically submitted to Internal Revenue Service (IRS) shortly after approval. The complete 2020 Form 990 package was provided to the full Board of Directors before it was filed with IRS.

Form 990, Part VI, Section B, Line 12C

Semi-annually, all members of the Board of Directors are required to review and sign a conflict-of-interest form disclosing any such issues. Forms are reviewed to ensure compliance, and any areas of concern are thoroughly discussed and remedied at the time, and throughout the year. Board members having a conflict of interest are required to recuse themselves from voting on such matters for which they have a conflict.

National Health Emergency: On January 30, 2020, the World Health Organization declared the outbreak of a respiratory disease caused by a new coronavirus as a pandemic. First identified in late 2019 and known now as COVID-19, the outbreak has impacted millions of individuals worldwide. In response, and to combat the outbreak, many countries have implemented stringent measures, which have impacted global business operations. In March, the United States declared a national emergency due to the viral pandemic. It, and declarations by various states, and the outbreak of the virus itself, will have far reaching social, economic, and financial impacts on the United States going forward. In 2020, COVID-19 affected Baltimore Under Ground Science Space, Inc.'s (BUGSS) operations as well as that of its suppliers, vendors, donors and other key stakeholders. For some months, BUGSS' one employee was furloughed and, comparing 2020 to 2019, membership revenue was the same but the amount collected for iGEM memberships was reduced by $1,000 and the amount from adult and youth courses jointly was reduced by $4,000. Although the financial statements reflect declines to certain balances and results from typical operating levels (cash, donations, pledges, grants, net assets, etc.) as a result of the public's response to COVID-19 in the last half of the tax year, the overall impact on the financial status and operations of the organization cannot be fully determined. Accordingly, no impairments were recorded as of the statement of financial position date (December 31, 2020), however, due to significant uncertainty surrounding the situation, management's judgment regarding this could change in the future. In addition, while the organization's financial condition, results of operations, and cash flows could be negatively impacted, the extent of the impact cannot be reasonably estimated at this time. Looking forward, the extent to which the pandemic impacts the organization's activities and results in tax year 2021 and beyond will depend on future developments, which are highly uncertain and cannot be predicted, including new information, which may emerge concerning the severity of the coronavirus and actions taken to contain the coronavirus and its impact, among others.

IRS990/Desc0Program Service 1, Community and Individual Member Projects: Central to BUGSS' mission is providing laboratory space to our community's individuals and groups, who wish to conduct experiments of their own design. Individuals pay memberships to BUGSS, which fees enable them to access the facilities and equipment of the laboratory as well as a small amount of disposable reagents. In this way, we dramatically reduce startup costs for those interested in pursuing high risk, non-traditional, multidisciplinary and early-stage projects. In 2020, the BUGSS laboratory supported two community projects. The first was an Inner Harbor Project in which eight BUGSS members performed DNA extraction, amplification and sequencing of water samples from Baltimore's Inner Harbor. Collaborating with scientists at Institute of Marine and Environmental Technology, these samples were analyzed to identify all of the organisms living throughout the Harbor community. The second project was Open Insulin. In addition to our community projects, we had individual members who pursued projects at BUGSS. These ranged from the production of plant cellulose in bacteria to the engineering of bacteria to support corals against environmental change, molecular cloning of genes needed for cancer therapeutics, and isolation of bacterial species with beneficial properties.

IRS990/ProgSrvcAccomActy2Grp/Desc0Program Service 2, iGEM: In 2020, BUGSS hosted, sponsored and mentored the Baltimore Bio-Crew, a team competing in the 2020 international Genetically Engineered Machines competition (iGEM). BUGSS provided laboratory space, supplies, and mentoring for the team throughout the spring and summer. In November, BUGSS also provided registration, travel and housing for four participants to attend the iGEM Giant Jamboree held at Hynes Convention Center in Boston. Our program accomplishments include the establishment of a team of eighteen high school students from Baltimore City and surrounding counties, and seven mentors who were professional scientists, postbaccalaureate and graduate students and postdoctoral fellows. They focused on designing and building genetically modified bacteria that expressed a protein to enhance the growth of beneficial gut bacteria. This system could be given to patients as a probiotic in conjunction with antibiotics, thereby enhancing the regrowth of beneficial bacteria in the gut and preventing opportunistic infections that often follow antibiotic use. In addition, they studied the potential for implementation of the device, and the project's ethical implications. The team earned a gold medal and the award for Best High School Measurement.

IRS990/ProgSrvcAccomActy3Grp/Desc0Program Service 3, Youth Courses and School Programs: BUGSS also offers courses to groups of students (school programs) both in partnership with other local nonprofits like MERIT Health Leadership Academy and by direct recruitment of student populations such as homeschooled students. BUGSS expanded its school programming by offering four programs for students: Proteostasis: The Complicated Life Of A Protein; Build An Ecosystem!; TinkerCad For Students; and Water You Going To Do About CO2? These programs provide a valuable resource and laboratory access to a student population that would otherwise be unable to conduct laboratory experiments at such a rigorous level.

IRS990/ProgSrvcAccomActyOtherGrp/Desc0Program Service 4, Adult Courses and Seminars: BUGSS teaches in-depth, hands-on laboratory courses to members and the general public. Courses explore aspects of modern biotechnology, bioinformatics, genetics and molecular biology, and focus on teaching participants both concepts and laboratory techniques. In addition to our in-depth courses, BUGSS offered a series of short, weekday-evening courses in basic molecular-biology laboratory skills. In 2020, BUGSS hosted seven courses, which ranged from two hours to multi-week courses of twelve hours total. Average enrollment in the courses was twelve students, ranging in age from teens to over 60. Courses included Introduction To Evolutionary Genetics, Sex Differences In The Brain, Genome Engineering And CRISPR, Seeing Learning And Memory Through The Microscope, Build-a-Genome, and Hunting For Genes: How Does A Genome Wide Association Study Work? Importantly, through our association with the Collaborative Teaching Fellows Program of Johns Hopkins University and the University of Maryland Schools of Medicine, four of the courses were taught by postdoctoral fellows and two courses were adaptations of the fellows' own research into a course suitable for the general public. Laboratory-skills courses were offered on a bi-weekly basis from January to March, and again in September. Topics ranged from plasmid DNA isolation to polymerase chain reaction, restriction digest, and gel electrophoresis. These served as an initial introduction to laboratory skills and techniques for those interested in building skills for the workplace or for learning techniques that could be used in subsequent projects.
