Uniform heavy SDS infection was achieved in the disease nursery and clear segregation of SDS resistance was observed in the mapping populations. Genomic DNA was extracted from the 243 lines and the three parents. Over 500 F4 seeds derived from crosses using E07080 as the SDS resistant parent were planted this summer. The plants from these seeds will be harvested this fall to form the mapping populations to map SDS resistance genes in E07080. For objective 2, Identify or confirm SDS resistance QTL present in advanced breeding lines entered in the SDS Regional Tests, cooperators report that advanced breeding lines tested each year in the SDS Regional Tests represent the best SDS resistant selections from all breeders participating in the tests. It is expected that multiple SDS resistance genes exist in these lines. We propose to use these lines to map or confirm SDS resistant QTL from multiple sources using a pedigree-based association mapping method. This year, 132 advanced breeding lines have been entered in the SDS Regional Test. Genomic DNA has been extracted from these 132 lines.Evaluation of soybean germplasm for new sources of resistance to the fungal pathogen Fusarium virguliforme, the cause of sudden death syndrome, Glen Hartman and Theresa Herman report eleven sets (nearly 1 per week) representing a total of 7,480 accessions have been evaluated to date in the first "tier" of evaluations. Of these 7,480 accessions, 3,961 are being advanced to the second tier of evaluation. Plans for next reporting period include the remainder of the accessions in the first tier evaluation (about 3,000) will be completed; the second tier testing will commence and is expected to be completed during this next quarter; and a tier three list of accessions will be compiled. Iron deficient chlorosis: Getting to the root of the problem; Iron deficiency chlorosis (IDC) is consistently rated as one of the most serious production problems affecting soybean yields in the upper Midwest. Project's Strategic Goal is to develop useful molecular markers that can identify IDC efficient genotypes and use state of the art genomic technologies to identify genes involved in IDC efficiency or inefficiency. The ultimate goal of the research is to isolate candidate genes and develop markers for these genes that will aid the soybean breeder in developing IDC resistant varieties. Screening for genetic resistance against soybean viruses; Previous research has found that the control of the disease caused by bean pod mottle virus (BPMV) could not be achieved through management of the virus vector, the bean leaf beetle. Therefore, the most practical and reliable method for disease control is disease resistance. However, adequate levels of resistance have not been available. Hill and Grau have developed a novel seed-based assay to measure the amount of virus in a seed to identify BPMV field tolerance. The method involves an engineered BPMV isolate that contains a gene for herbicide resistance. When soybean plants are inoculated with the engineered BPMV isolate, one can easily and rapidly assay for BPMV resistance. The location has a history of soybean and corn production. Plots were established, infested with F. virguliforme, and planted in early May. Supplies have been purchased to set up an irrigation system in late June or early July. To date this season we have received above average rainfall, and the plants in the plots are growing well. A few plants (0.1%) are showing early symptoms of SDS, which indicates that environmental and biological conditions have been suitable for infection. For objective two (Study SDS partial resistance in early maturing genetic material and provide resistant germplasm adapted to Minnesota for public and private breeders) that the soybean entries planted in the new field research location are early maturing varieties (maturity groups 1.0 to 1.9). Many are from Jim Orf's breeding program at the University of Minnesota, and others are from other breeding programs. The level of SDS that develops in these entries will be used to investigate partial resistance to SDS. For objective three (Determine characteristics of the pathogen that influence survival, spread and infection of soybean) that studies have been initiated in growth chambers and a greenhouse to investigate potential differences in virulence/aggressiveness among a group of isolates of F. virguliforme from MN and to determine environmental factors that influence survival of this pathogen. Molecular dissection of soybean aphid biotypes and aphid resistant soybean genes for controlling soybean aphids; The soybean aphid, Aphis glycines, is the most destructive soybean insect in the North-Central Region. "Provide a forum for state Extension specialists to coordinate research and Extension educational activities. Development of a high resolution genetic map for the soybean cyst nematode; Soybean cyst nematodes (SCN) can be managed using natural sources of resistance if the source of resistance can be matched to the nematode population. The reason for this "matching problem" is due to the fact that SCN populations adapt and reproduce on resistant soybean plants. Nematodes that can grow on resistant plants are referred to as virulent. The consequence of this "selection for virulent SCN biotypes" is that SCN resistance becomes less effective over time and new types of resistant plants must be deployed. It is time consuming and expensive to test SCN populations for virulence using greenhouse assays, thus a rapid DNA-based test is needed to detect the extent of virulent biotypes that have built up in nematode populations in the field. Our group has devised a simple quantitative real-time PCR assay that can detect SCN genes in field samples quickly and inexpensively (it costs $1/sample and takes 2 hours). One critical component we are missing is a dense genetic map for SCN that defines the exact region in the genome containing SCN virulence genes. This DNA sequencing project will allow most sequence differences between the avirulent and virulent lines to be identified. These sequence differences, in turn, will be used to map-base clone SCN virulence genes. The virulence genes will then be used to devise a rapid SCN virulence test, which will allow soybean fields to be monitored for virulent nematode build up. Such information will in turn allow the most effective SCN resistant soybean to be grown, thus minimizing the damage caused by SCN.The goal of this project was to use the very deep genomic DNA sequence data for two soybean cyst nematode (SCN) biotypes differing in virulence to identify 1500 SNPs and to map them to the SCN genetic map. In this project we plan use a SCN reference sequence generated by the Joint Genome Institute (JGI) and re-sequence data generated by a new DNA sequencing technology being commercialized by Applied Biosystems (ABI) called SOLiD (sequencing by oligonucleotide ligation and detection). |
